In this study, we have evaluated changes in immunoglobulin concentration and cytokine expression by different sensitization methods, for producing food allergy rat models. 4-week-old male BN rats were divided into four groups. 1) control group, 2) administration intraperitoneally of 0.5 mg ovalbumin (OVA) once a week, 3) administeration orally of 1 mg OVA daily, 4) administration orally of 2 mg OVA every 2 days. After 35 days, plasma OVA-specific IgE levels were found to be increased significantly only in the intraperitoneal group, indicating sensitization to OVA. In addition, the expression level of IL-4/IL-2 in the spleen was elevated in the intraperitoneally administered group. This suggests that the Th1/Th2 balance may be tilted toward Th2. This may have resulted in excessive humoral immunity and increased OVA-specific IgE. On the other hand, oral administration of allergens may induce oral tolerance, our results suggest that sensitization by intraperitoneal administration may be more
